Mikroba (Bakteri) Lingkungan

Tujuan : Untuk mengetahui macam jenis bakteri yang ada di lingkungan (air, tanah, udara)

Dasar Teori : 
Kondisi lingkungan yang mendukung dapat memacu pertumbuhan dan reproduksi bakteri. Faktor-faktor lingkungan yang berpengaruh terhadap pertumbuhan dan reproduksi bakteri adalah suhu, kelembapan, dan cahaya.

Alat + Bahan :  Agar nutrien
                        Cawan Petri
                        Tabung Reaksi
                        Ose

Sampel          :  Air, Udara , Tanah

Cara Kerja    :

I. PEMBUATAN MEDIA 
  • Masukkan R/12,75 agar nutrien 
  • Lalua encerkan dalam 500mL

II. ISOLASI

     1. Udara
         Diamkan agar di tempat tertentu, lalu buka cawan dan diamkan terbuka selama 15 - 20 menit inkubasi
         selama 18 - 24jam dalam inkubator

     2. Tanah
          Masukan 1gr tanah ke dalam tabung reaksi yang telah berisi aquadest streril aduk lalu tanam ke media
          inkubasi selama 18 - 24jam dalam inkubator

     3. Air
         Ambil air yang di curigai , lalu tanam ke media dengan menggunakan ose inkubasi selama 18 - 24jam
         dalam inkubator
