 Pengenalan Bentuk Normal Pada Bahan Pemeriksaan Tepung


Tujuan :
Untuk mengetahui struktur dan morfologi dari tepung

Alat :
·         Objek glass
·         Cover glass
·         Lidi
·         Tissue
·         Pipet pastuare

Bahan :
·         Tepung terigu
·         Tepung maisena
·         Tepung beras
·         Tepung kanji
·         Tepung hongwe
·         Kentang

Reagen :
·         NaCl 0,85%
·         Eosin 2%
·         Lugol 2%

Prosedur kerja :
·         Menyiapkan semua peralatan dan bahan yang diperlukan
·         Menyiapkan objek glass untuk masing – masing tepung
·         Membersihkan objek glassndengan alkohol atau dengan cara flamming agar lemaknya hilang
·         Mengambil 1 lidi dan mengambil 1 tetes tepung dan di letakkan pada obhek glass, masing – masing 1 tetes tepung dan 1 tetes reagen, kemuadian di aduk rata dan ditutup dengan cover glass (jangan sampai ada gelembung)
·         Memeriksa di bawah mikroskop dengan perbesaran 10x dan untuk memperjelas hunakan perbesaran 40x , kemudian amati berupa :
§  Bentuk  : kotak / bulat / lonjong
§  Tepi      : halus / bergerigi
§  Sifat      : menyerap warna / tidak
§  Struktur didalamnya : berserat / lonjong / berlapis – lapis
